INGENIEUR.E CHEF.FE DE PROJET EN DEVELOPPEMENT TECHNOLOGIQUE ET EXPERIMENTAL CDD



Au sein de la plateforme TherA-Tech et sous la responsabilité de son directeur, vous serez en charge du pilotage et de la conduite de projets de développements technologiques expérimentaux, incluant la conception et la réalisation de protocoles expérimentaux, de la coordination du suivi technique de l'infrastructure, de la gestion du budget et des ressources allouées. A ce titre, vous coordonnerez une équipe d’ingénieurs et techniciens.

Vos activités principales :
- Étudier les besoins scientifiques, techniques et expérimentaux, proposer des méthodes de mesure, de caractérisation ou d'observations adaptées et établir des offres de prestation
- Analyser les contraintes métrologiques et concevoir ou faire évoluer la chaîne d'expérimentation et de mesure
- Concevoir des développements technologiques mutualisés et innovants, en relation avec les projets des utilisateurs ou partenaires, et établir le cahier des charges
- Organiser le suivi des réalisations, valider et qualifier les dispositifs ou les méthodes aux différentes étapes
- Conseiller les utilisateurs et les partenaires sur les possibilités et limites des techniques disponibles, sur l'interprétation des données
- Assurer une veille technologique
- Piloter la réalisation des mesures, les interpréter et les valider
- Gérer et encadrer l'ensemble des ressources humaines, techniques et financières allouées aux expériences et à la plateforme
- Mettre en place les procédures de traçabilité et d’assurance qualité
- Participer à la rédaction de dossiers dans le cadre des demandes de financement
- Promouvoir les services de la plateforme et prospecter
- Présenter, diffuser et valoriser les réalisations

PRECLINICAL IMAGING RESEARCH SCIENTIST (M/F) LABORATORY MANAGER

Activities:

Thèse sur la conception d’un agent radiomarquable universel sélectif pour l’imagerie TEP et le traitement par RTIV

L'imagerie moléculaire, en particulier l’imagerie de tomographie par émission de positons (β+) (TEP) est devenue indispensable au diagnostic de nombreuses maladies comme les cancers. L'essor de cette technique non invasive est lié à l’obtention d’images tridimensionnelles, après injection d’un radiotraceur, permettant de localiser et quantifier des cibles spécifiques à la pathologie. Les radiotraceurs sont également employés pour le traitement de cancer par radiothérapie interne vectorisée (RTIV). Ces radiotraceurs associent majoritairement un agent de ciblage, interagissant avec un biomarqueur de la pathologie, et un agent chélatant complexant un radionucléide permettant l'enregistrement d'images. Récemment une nouvelle technique de radiomarquage au fluor-18 par chimie de complexation (i.e., formation du complexe d'aluminium-fluor-18) a vu le jour et est devenue une technique de choix pour le développement de nouveaux radiotraceurs. En fonction de la nature des agents chélatants, différents radionucléides peuvent être complexés par les traceurs. Le fluor-18 (18F) et le gallium-68 (68Ga) sont des radionucléides de choix pour l'imagerie TEP et le lutétium-177 (177Lu) pour la RTIV, mais ces trois radionucléides possèdent des propriétés de complexation différentes. Ainsi, l’aluminium-fluor-18 peut être complexé par des agents chélatants de types HBED-CC et NOTA, le gallium-68, par des agents chélatant de types HBED-CC, NOTA et DOTA et le lutécium-177, par des agents chélatant de type DOTA.

L’objectif de ces travaux de thèse est d'optimiser la structure chimique d'un nouvel agent chélatant bifonctionnel (ACB), développé au cours d'une thèse précédente, pouvant être (i) radiomarqué à l’aluminium-fluor-18, au gallium-68et au lutécium-177 par chimie de complexation, et (ii) couplé à des peptides de ciblage par bioconjugaison. Le but est de préparer des « peptides-chélatants » pouvant être utilisés en imagerie TEP et en RTIV pour une application en théranostic (thérapie + diagnostic) s’insérant dans une démarche de médecine personnalisée.

Methodological developments in micro-MRI and NMR spectroscopy

MISSION
The engineer will conduct research in MRI, principally at 11.7T on an NMR spectrometer equipped
with a micro-imaging probe:
- for the in vitro and in vivo study of animal models such as Zebrafish, complex cellular systems (organoids,
3D cultures) or for the ex vivo study of fresh or fixed tissues
- for the study of bio sourced gels based on polysaccharids (starch, arabinoxylanes) or protein (corn zein) in order to conceive bio polymer matrices with controlled functionalities allowing, for example, the liberation of active principles initially contained in these matrices. This research work contains several interconnected contributions:
WP1: Elaboration of experimental set-ups adapted to the study of multicellular systems, and zebrafish larvae.
WP2: Validation of the sequences adapted to the study of multicellular systems, zebrafish larvae and tissues.
Adaptation of the protocols for the in vivo imaging of the zebrafish.
WP3: Optimization of the study of dynamic monitoring of functional bio sourced matrices. Implementation of the study of dynamic monitoring of functional bio sourced matrices.
The candidate will need an ability to imagine and conceive experimental plans adapted to the study of multicellular systems, zebrafish larvae (WP1) and hydrogels whose shape/form evolves through time (WP3). The candidate will carry the realization of the experiments with the MRI (WP2 and WP3), their interpretation and traceability, from data to analysis reports, and their valorization in communications in conferences and articles in scientific journals.

Développements méthodologiques en micro-IRM et en spectroscopie par RMN

Description du service et place de l’agent dans l’organisation :

L’ingénieur(e) de recherche recruté(e) sera rattaché(e) à l’UAR Biosit qui gère 14 plateformes dont la Plateforme de Recherche en Imagerie et Spectroscopie Multimodales PRISM, (https://www.pf-prism.org/) qui propose des solutions d'imagerie et de spectroscopie pour une large gamme d’applications, allant du secteur biomédical jusqu’à l’agroalimentaire en passant par la chimie. PRISM dispose de 4 imageurs par Résonance Magnétique (IRM 1.5T, 4.7T, 7T and 11.7T), 2 spectromètres de RMN à haut champ (2 x 500MHz) et 6 relaxomètres (0.35T et 0.47T). PRISM fait partie du réseau des plateformes technologiques du Grand Ouest en sciences du vivant et de l’environnement Biogenouest (https://www.biogenouest.org/) et de l’infrastructure nationale de recherche “France Life imaging”. PRISM est certifié ISO 9001 : v2015.

Le (la) candidat(e) intégrera la plateforme PRISM, en particulier la composante Bio-SCANs sur le site de
Villejean incluant de fortes interactions avec la composante Agro-SCANs de Beauregard (UR OPAALE,
INRAE). Il (elle) travaillera dans le cadre d’un projet dont l’objectif est de développer de nouvelles méthodes
en IRM et en spectroscopie de RMN de façon parallèle, sur la sonde de micro-imagerie de la plateforme
PRISM. Cette double approche bénéficiera de l’expertise et du savoir-faire déjà existants sur la plateforme
des deux modalités (imagerie et spectroscopie) dans le domaine du médical (Bio-SCANs) et de la science
des matériaux (Bio-RMN et Agro-SCANs).

Missions et activités:
Mission : L’ingénieur(e) mènera des recherches en IRM principalement à 11,7T sur un spectromètre de RMN équipé d’une sonde de micro-imagerie :
- pour l’étude in vitro et in vivo de modèles animaux de type Zebrafish, de systèmes cellulaires complexes (organoïdes, cultures 3D) ou pour l'étude ex vivo de tissus frais ou fixés ;
- pour l’étude de gels biosourcés à base de polysaccharides (amidon, arabinoxylanes) ou d’une protéine (zéine de maïs) dans l’optique de concevoir des matrices de biopolymères à fonctionnalités contrôlées permettant par exemple la libération de principes actifs pharmaceutiques initialement encapsulés dans ces matrices.

Ce travail de recherche comporte plusieurs contributions, qui s’auto-nourrissent :
WP1 : Mise au point de dispositifs expérimentaux adaptés à l’étude des systèmes multicellulaires, et de larves de zebrafish.
WP2 : Validation des séquences adaptées à l’étude des systèmes multicellulaires, de larves de zebrafish et de tissus. Adaptation des protocoles pour l’imagerie in vivo des zebrafish.
WP3 : Optimisation des études de suivi dynamique des matrices biosourcées fonctionnelles. Mise en œuvre des études de suivi dynamique des matrices biosourcées fonctionnelles.

Le(la) candidat(e) aura une capacité à imaginer et concevoir des dispositifs expérimentaux adaptés à l’étude des systèmes multicellulaires, de larves de zebrafish (WP1) et d’hydrogels dont la forme évolue dans le temps (WP3). Il(elle) portera la réalisation des expérimentations en IRM du WP2 et WP3, leur interprétation et leur traçabilité, de la donnée à l’analyse sous forme de rapports, et leur valorisation sous forme de communications dans des congrès et d’articles dans des revues scientifiques.

Post-doctoral position in Medical Image Processing (18 months)

The Computer-Assisted Medical Interventions (CAMI) team of the TIMC laboratory (Grenoble, France) is
looking for a highly motivated postdoctoral candidate with a strong background in medical image processing
(registration and segmentation) and experience in deep learning approaches with a focus on ultrasound imaging.
The candidate will work on the development of a multimodal deep learning registration method between pre-
operative 3D MR images and intraoperative 3D transrectal ultrasound images. This method should be able to
provide a deformation eld to link the two images. The candidate will be expected to publish his/her research
in leading international journals and conferences in the eld, participate in the supervision of PhD students and
contribute to the collaborative projects within the laboratory.
The TIMC laboratory is located minutes away from the Grenoble University Hospital, allowing for a close and
effective collaboration with clinical & industrial partners. Grenoble is a welcoming student city at the heart of
the French Alps.
DURATION : 18 months (as soon as possible from 01/2023)

Post-Doctoral Position on Development of a 3D platform for Passive Acoustic Mapping of cavitation

CONTEXT
Therapeutic ultrasound offers great perspectives for minimally invasive surgery, enhanced drug delivery or cancer immunotherapy. It now addresses an extensive range of indications, from prostate or brain tumors to glaucoma. Among other mechanisms, various emerging applications rely on the phenomenon of ultrasound cavitation, which represents the oscillation of ultrasound-induced microbubbles. In any of these applications, monitoring the treatment in real-time is required for potential clinical applications. While active ultrasound B-mode imaging is well suited to monitor thermal or mechanical permanent alteration of tissues, the microbubble activity – directly actuated by high-intensity ultrasound – cannot be characterized in real-time using an active ultrasound scanner because of dazzling effects.

To localize and quantify the cavitation activity, Passive Acoustic Mapping (PAM) beamforming techniques have been used, but they suffer from weak spatial resolution in the axial direction, perpendicular to the array, when conventional imaging arrays are used. This is even more critical for applications requiring a 3D monitoring of the cavitation activity. Indeed, even if a few works have demonstrated the feasibility of 3D passive acoustic mapping of cavitation, especially in the context of transcranial therapy with very specific hemispherical arrays, 3D mapping with conventional matrix arrays suffers from very low axial resolutions due to diffraction effects associated to the low apertures of the probes. Adaptive beamformers have proven to be an effective way to enhance PAM resolution but do not entirely compensate for those diffraction effects and complimentary research avenues such as dual array mapping have to be considered to achieve millimetric or sub-millimetric resolutions in any direction.

MISSION

Within the CAVIIAR ANR Project framework, the candidate will have to evaluate the feasibility of a dual-probe
mapping based on two commercial matrix arrays with electromagnetic tracking of their positions. He will then develop a 3D navigation and imaging platform combining one or two open ultrasound scanners connected to two matrix arrays, a tracking unit giving translational and rotational data of each probe, a 7-axis robot that will give reference positions of the probes in an evaluation step, and a 3D navigation station providing tools for communication with other devices, reconstruction and visualization in a 3D virtual environment with fusion capability of active and passive US images. The platform will aim at providing dual-array 3D PAM of cavitation events triggered in agar-based phantoms with millimetric or sub-millimetric resolution in any direction using two commercial matrix-array probes.

L'Inserm en partenariat avec Sorbonne Unviersité propose au sein du Laboratoire d'Imagerie Biomédicale (LIB, U1146) un poste de chaire de professeur.e junior en Innovation en neurotechnologies pour une meilleure probabilité de dispositifs médicaux.
